The academic calendar shows the deadlines and scheduled events that are relevant to students, teaching and technical-administrative staff of the University. Public holidays and University closures are also indicated. The academic year normally begins on 1 October each year and ends on 30 September of the following year.

Learning outcomes
Il corso si prefigge di fornire le conoscenze necessarie alla comprensione delle potenziali applicazioni dei microrganismi in campo agroalimentare, industriale e ambientale. Fornire un’ampia visione della biodiversità metabolica dei microrganismi in natura e la loro potenzialità di produrre particolari metaboliti. Utilizzo delle tecniche del DNA ricombinante per lo sviluppo di ceppi geneticamente modificati.
Program
1. Introduzione al corso. Discipline afferenti alle Biotecnologie Microbiche. Evoluzione delle Biotecnologie. Operazioni biotecnologiche.
2. Diversità microbica: benefici ambientali ed economici. Microrganismi e gruppi filogenetici più rappresentativi. Metodi di valutazione della biodiversità e detection di procarioti non coltivabili.
3. Metagenomica. Analisi delle librerie metagenomiche: approccio funzionale e basato sulle sequenze. Esempi di outcome della metagenomica.
4. Strategie per l’isolamento selettivo, la caratterizzazione e la selezione di nuovi microrganismi di interesse biotecnologico. Un caso studio: isolamento di batteri oligotrofici da ecosistema marino. Procedure per la caratterizzazione microbica: il sistema polifasico. Conservazione dei microrganismi ex situ ed in situ.
5. Enzimi microbici. Requisiti dei microrganismi produttori. Produzione industriale. Bioreattori e chemostati. Enzimi immobilizzati. Glucosio isomerasi. Alfa amilasi. Pullulanasi. Enzimi proteolitici. Subtilisine. Chimosina.
6. Gli antibiotici. Classi di antibiotici: β-lattamici (penicilline e cefalosporine), aminoglicosidi, macrolidi e tetracicline. Misura dell’attività antibiotica: MIC, saggi in piastra. Antibiotico-resistenza. Sintesi microbica di antibiotici.
7. Insetticidi microbici. Cristalli parasporali di Bacillus thuringensis e B. sphaericus. Attività insetticida di B. popilliae, Baculovirus, Photorhabdus luminiscens e Xenorhabdus nemathophilus. 8. Sintesi microbica di biopolimeri (xantani e poli-idrossialcanoati). Applicazioni industriali dello xantano. Plastiche biodegradabili.
9. Vaccini a sub-unità, ricombinanti e sintetici. Uso di batteri GRAS come veicolo di vaccini.
10. Ingegneria metabolica in Saccharomyces cerevisiae. Esempi di obiettivi metabolici: estensione del substrato utilizzabile; miglioramento della produttività e resa; riduzione dei sottoprodotti; miglioramento della resa del processo; estensione della produzione, compresa la produzione di proteine eterologhe.
11. Produzione di biosurfattanti e loro applicazioni commerciali.

La prova d’esame consisterà in una valutazione orale del grado di apprendimento conseguito sul programma teorico svolto.
