Learning outcomes
The course introduces students to enzymology and the application of enzymes in medical biotechnology and medicine. The students will acquire advanced knowledge on enzymes mechanisms and regulation, technology of enzyme production, new biotechnological strategies in engineering of enzymes aimed to their improved use in the medical and pharmaceutical fields. The main part of the course will be focused on selected applications of enzymes in biomedicine, diagnostics and therapy.

Theory
• Introduction to enzymes and enzyme kinetics
• Production of recombinant enzymes/proteins
• Protein engineering: directed and rationale evolution for production of new enzymes
• Enzyme therapy in metabolic disorders:
1. Lysosomal storage diseases
2. Disorders of amino acid metabolism
3. Disorders of nucleotide metabolism
• Enzymes in cancer therapy
• Fibrinolytic enzymes for thrombolytic therapy
• Enzymes in diagnosis and diagnostics
Laboratory
Session1: purification of recombinant wild type cystathionine beta synthase and its pathogenic variants via affinity chromatography. Analysis of the affinity purified enzyme samples by SDS-PAGE.
Session2: characterization of wild type and pathogenic mutants using kinetic and spectrophotometric methods (cofactor content of the enzyme and steady-state kinetic properties)
