Learning outcomes

The course aims to make students understand the basic stages of the birth of modern biotechnologies and the current applications of recombinant biotechnology in biomedical and pharmaceutical fields. We will discuss issues of a scientific, ethical and biological nature. The course also aims to provide an updated view of recombinant techniques including techniques for reproductive and therapeutic cloning, technic for the reproduction of recombinant drugs.

Program

1) History of modern Biotecnology. The Reproductive cloning. The Therapeutic cloning. Critical review of two fundamental scientific works: 1) Wilmut I, Schnieke AE, McWhir J, Kind AJ, Campbell KH.Viable offspring derived from fetal and adult mammalian cells.Nature. 1997 Feb 27;385(6619):810-3.
2) Schwartz SD, Regillo CD, Lam BL, Eliott D, Rosenfeld PJ, Gregori NZ, Hubschman JP, Davis JL, Heilwell G, Spirn M, Maguire J, Gay R, Bateman J, Ostrick RM5, Morris D, Vincent M, Anglade E, Del Priore LV, Lanza R. Human embryonic stem cell-derived retinal pigment epithelium in patients with age-related macular degeneration and Stargardt's macular dystrophy: follow-up of two open-label phase 1/2 studies. 2) Cloning vectors. Role of DNA ligase restriction enzymes, alkaline phosphatase. Plasmids. Methods for introducing DNA into a host cell: transformation and transfection. Vectors selection and isolation.

4) Clinical applications of recombinant drugs. Human recombinant TSH. production technique, commercial products, pharmacokinetic properties, approval for clinical use. Use in the diagnosis and follow-up of differentiated thyroid carcinoma
5) Clinical applications of recombinant drugs. Recombinant erythropoietin: production technique, commercial products, pharmacokinetic properties, approval for clinical use. Recombinant insulin: production technique, pharmacokinetic properties, clinical use, commercial products.
